چکیده

Recently great advances have been made in the preparation of pure myoglobin by the introduction of ion exchange cellulose column chromatography (1, 2). Myoglobin, however, shows a strong tendency to be oxidized to metmyoglobin during isolation procedures. For this reason all myoglobin preparations reported earlier were obtained in the form of metmyoglobin. Therefore, oxymyoglobin has usually been obtained from metmyoglobin by reduction with sodium hydrosulfite, a procedure which has remained questionable especially from the point of view of its physiological activity. This communication deals with procedures for the isolation of native oxymyoglobin from fresh horse heart, and the chromatographic separation of oxymyoglobin from metmyoglobin with diethylaminoethyl cellulose.

Decrease in oxygen affinity of myoglobin by formylation of vinyl groups of heme.

Three kinds of green synthetic myoglobin were prepared by recombination of horse heart apomyoglobin with spirographis (2-formyl-4-vinyl-), isospirographis (2-vinyl-4-formyl-), and 2,4-diformyldeuterohemins. The optical and oxygen binding properties of the reconstituted myoglobins containing two isomeric monoformyl-monovinylhemins were found to be different. The oxygen affinities (P50) of spirog...

The absorption spectra and extinction coefficients of myoglobin.

In a study of the quantitative variations of myoglobin in muscles, the need arose for reliable absorption spectra and extinction coefficients of derivatives of both myoglobin and hemoglobin in the visible range. Sufficient data on hemoglobin are available in the literature, but those for myoglobin (1, 2) are conflicting and suggest the need for further study. Therefore, it seemed advisable to p...

Seaweed Polysaccharides (Laminarin and Fucoidan) as Functional Ingredients in Pork Meat: An Evaluation of Anti-Oxidative Potential, Thermal Stability and Bioaccessibility

The anti-oxidative potential of laminarin (L), fucoidan (F) and an L/F seaweed extract was measured using the DPPH free radical scavenging assay, in 25% pork (longissimus thoracis et lumborum (LTL)) homogenates (TBARS) (3 and 6 mg/mL) and in horse heart oxymyoglobin (OxyMb) (0.1 and 1 mg/mL). The DPPH activity of fresh and cooked minced LTL containing L (100 mg/g; L100), F100 and L/F100,300, an...

The Molecular Weight of Crystalline Catalase.

In 1933 Stern (2) used the diffusion method of Northrop and Anson (3) to determine the molecular weights of purified catalase from horse liver, pig liver, cow liver, horse kidney, and cow erythrocytes. He found the average value for the molecular weight of catalase to be 68,900, and stated that the catalase molecule has a dimension similar to that of hemoglobin. In 1938 Stern and Wyckoff (4) pu...

Isolation of sperm whale myoglobin by low temperature fractionation with ethanol and metallic ions.

1. A method is described for the isolation and purification of myoglobin from sperm whale skeletal muscle. The aqueous extract is brought to 25 % ethanol by volume at 10 to lZ” and pH 6.5. A fraction precipitated with lead subacetate is discarded. Myoglobin is precipitated from the supernatant solution at pH 7.20 by adding zinc acetate to a concentration of approximately 10 InM. The precipitate...
